高效液相色谱—串联质谱法同时测定蜂胶中19种酚类化合物

本实验建立一种能够同时测定蜂胶中19种酚类化合物的高效液相色谱 串联质谱法(HPLC-MS-MS)的检测方法。采用85%的乙醇水对粉碎混匀后的蜂胶样品进行提取,样液经Waters T3色谱柱分离,并使用流动相(甲醇和0。1%甲酸水溶液)进行梯度洗脱,采用电喷雾离子化、多反应监测模式进行检测,外标法定量。结果表明:19种酚类化合物在线性范围内的线性关系良好,相关系数(R2)均大于0。99,回收率为87。4%~107。8%,相对标准偏差(RSD)为0。9%~7。6%。该方法具有前处理操作简单、定性定量准确等特点,可用于蜂胶样品中19种酚类化合物的检测,为蜂胶质量评价提供了技术支撑。
Simultaneous determination of 19 phenolic compounds in propolis by high—performance liquid chromatography—tandem mass spectrometry
To develop a high-performance liquid chromatography-tandem mass spectrome-try method for the simultaneous determination of 19 phenolic compounds in propolis.Propolis samples were extracted by 85%ethanol after crushing and mixing,and the sample solution was separated on a Waters T3 column with a gradient elution using the mobile phase(methanol and 0.1%formic acid aqueous solution).The detection of phenolic compounds was performed by elec-trospray ionization(ESI)and multiple reactions monitoring(MRM)modes,and quantified by an external standard method.The 19 phenolic compounds showed good linearity in the linear range with the correlation coefficients(R2)greater than 0.99.The recoveries ranged from 87.4%to 107.8%with the relative standard deviation(RSD)of 0.9%to 7.6%.The method is simple and efficient for analysis of 19 phenolic compounds in propolis,and can provide technical supports for the quality evaluation of propolis.
