Study on optimization of extraction process of Gloeostereum incarnatum antioxidant peptide by probiotic fermentation and its anti-exercise fatigue effects
Using Gloeostereum incarnatum as raw material,the antioxidant peptide was extracted by pro-biotic fermentation.Using ABTS free radical scavenging rate as an indicator,probiotic was screened and precipitation was optinized.On the basis of single factor experiments,response surface methodology was used to optimize the process of extracting antioxidant peptides from Gloeostereum incarnatum by probiotic fermentation,and the anti fatigue effect of Gloeostereum incarnatum antioxidant peptides on exercise rats were studied.The results showed that the optimal fermentation conditions for preparing Gloeostereum incar-natum antioxidant peptides were Bacillus subtilis as probiotics,precipitation pH 6,liquid to material ratio 4.0∶1(ml/g),fermentation temperature 34 ℃,dosage of Gloeostereum incarnatum 53 g,fermentation time 24 h.Under these conditions,scavenging rate of Gloeostereum incarnatum antioxidant peptides on the ABTS free radical was 93.44%.The antioxidant capacity test showed that the activity of Gloeostere-um incarnatum antioxidant peptides was best when its molecular weight was less than 3 kDa.The antifa-tigue test of exercise rats showed that gavage of Gloeostereum incarnatum antioxidant peptides could im-prove endurance and anti-fatigue ability of rats.
