天麻益生菌复配对改善小鼠睡眠作用的影响

探究天麻益生菌复配改善小鼠的睡眠作用及机制.将70只Balb/c小鼠随机分成空白对照组、阳性药组、天麻乙醇提取物低剂量组、天麻乙醇提取物高剂量组、益生菌组、低剂量天麻益生菌复配组和高剂量天麻益生菌复配组.通过小鼠自主活动试验、戊巴比妥钠阈上及阈下剂量催眠试验,测定小鼠脑组织中γ-氨基丁酸(GABA)、多巴胺(DA)和血清中去甲肾上腺素(NA)的含量,以评价其改善睡眠的作用.结果显示:高剂量天麻益生菌复配组能有效抑制小鼠的自主活动,提高阈下剂量的小鼠入睡率,缩短阈上剂量的小鼠入睡潜伏期及延长睡眠时间,提高小鼠脑组织中GABA含量,降低脑组织中DA含量及血清中NA含量.表明高剂量天麻乙醇提取物与益生菌复配后能改善小鼠的睡眠质量.
The effect and mechanism of Gastrodia elata probiotics compound on sleep improvement in mice were explored.A total of 70 Balb/c mice were randomly divided into blank control group,positive drug group,low dose of Gastrodia elata ethanol extract group,high dose of Gastrodia elata ethanol extract group,probiotics group,low dose Gastrodia elata combined with probiotics group and high dose Gastro-dia elata combined with probiotics group.The sleep-improving effect was evaluated by the mice voluntary activity test,the upper and subthreshold dose of pentobarbital sodium hypnotic test,and the levels of γ-aminobutyric acid(GABA)content and dopamine(DA)content in mice brain tissue,and norepineph-rine(NA)in serum.The results showed that the high dose of Gastrodia elata combined with probiotics could significantly inhibit the autonomic activity of mice,increase the sleep onset rate of mice at the sub-threshold dose,shorten the sleep onset latency and prolong the sleep duration of mice at the suprathresh-old dose,increase the content of GABA in mice brain tissue,and reduce the content of DA in mice brain tissue and NA in serum.In conclusion,the combination of high dose Gastrodia elata ethanol extract and probiotics could improve the sleep quality of mice.
