The components of Lophatherum gracile Brongn.flavonoids(LGF)were analyzed,and the improvement effects of LGF on cardiomyocyte injury induced by angiotensin(Ang Ⅱ)were investigated.The results showed that LGF was mainly composed of isoorientin,chlorogenic acid,tricin,orientin,iso-vitexin,rutinoside,cynaroside,quercetin and vitexin,with a molar ratio of 3.36∶4.22∶1.15∶20.59∶17.99∶13.19∶2.54∶1∶7.97.Compared with M-Ang Ⅱ,the surface area of cardiomyocyte in the high(5 mg/mL),medium(2.5 mg/mL),and low(1 mg/mL)dose LGF groups were significantly reduced.The contents of malondialdehyde(MDA)in cardiomyocytes were significantly reduced by 77.54%,57.85%,and 48.31%,respectively.The antioxidant enzyme activities(SOD,CAT,GSH Px)were significantly increased,showing a dose-dependent relationship.The analysis of cardiomyocyte expression showed that compared with M-Ang Ⅱ,the mRNA relative expression levels of pro-apoptotic factors(Bax,Caspase-3,p53)in cardiomyocytes of high,medium,and low dose LGF groups were significantly or ex-tremely significantly reduced,while the mRNA relative expression level of anti-apoptotic factor Bcl-2 was extremely significantly increased.Overall,it showed that LGF could improve Ang Ⅱ-induced cardiomyo-cyte injury,and its mechanism of action may be related to reducing cellular oxidative stress levels and in-hibiting cell apoptosis.
