摘要
S-Klotho蛋白(S-Klotho)水平已被发现可以作为许多衰老相关疾病的进程指标,其水平降低与多个系统器官病理性病变及机体生理老化有关.近年来关于S-Klotho及其基因与各类衰老性疾病的相关研究越来越多,但外源注射S-Klotho干预疾病进程的研究还相对欠缺.本文就S-Klotho水平与人类疾病发展恶化以及全因死亡率的相关性、Klotho基因多态性与衰老性疾病的相关性、Klotho基因启动子部分甲基化与衰老性疾病进展的相关性以及外源注射S-Klotho干预疾病进程的临床前研究进行综述,以寻求该基因在衰老相关性疾病中所起作用的线索.
Abstract
The Soluble α-Klotho(S-Klotho)protein levels has been identified as a reliable means of assessing the advancement of many age-related ailments,and its decrease is related to pathological alterations of multiple or-gan systems and physiological aging of the body.In recent years,a growing body of research has investigated the correlation between S-Klotho plus its corresponding gene and various age-related ailments.Nevertheless,there is a shortage of evidence about the intervention of disease development by the exogenous administration of S-Klotho.This research provides an overview of the correlation between levels of S-Klotho and the onset and progression of hu-man diseases,the connection between Klotho gene polymorphism and age-related diseases,the association between partial methylation of the Klotho gene promoter and the advancement of age-related diseases,and preclinical inves-tigations on the potential therapeutic effects of exogenous administration of S-Klotho in halting disease progression so as to gain insights into the involvement of this gene in age-related diseases.
