Platelet promotes the epithelial-mesenchymal transformation process in lung cancer A549 cells and its mechanism
Objective To explore the effect of platelets(PLT)on the epithelial-mesenchymal transformation(EMT)of A549 cells and its mechanism.Methods PLT was isolated from healthy human peripheral blood and co-cultured with A549 cells with subse-quent microscopic observation of morphological changes in A549 cells.Western Blot was used to detect the expression of E-cadherin,N-cadherin,p-AKT,p-GSK3β and Snail.A total of 160 patients with lung cancer in our hospital were selected as the study group and 160 healthy people from the same period were collected as the control group.Peripheral blood PLT levels were compared between the two groups and the correlation between PLT levels of lung cancer patients and distant metastasis was analyzed.Results Co-culturing A549 lung cancer cells with PLT led to a spindle-shaped morphology,resembling that of mesenchymal cells.Western Blot revealed a decrease in E-cadherin expression and a significant increase in N-cadherin,p-AKT,p-GSK3β,and Snail expression after 48h co-culture.The PLT levels in lung cancer group were higher than those in control group(P<0.01).Furthermore,PLT levels were higher in the group with distant metastasis compared to the non-metastatic group(P<0.01).Conclusion PLT can promote EMT transformation in A549 cells,and increas the expression of p-AKT,p-GSK3β and Snail.PLT level was closely related to distant metastasis in lung cancer pa-tients.
