The Bacterial characteristics of intestinal contents in mice with Ganqichengpi diarrhea syndrome
Objective To explore the bacterial characteristics of intestinal contents in mice with Ganqichengpi diarrhea syn-drome,and to provide experimental evidence for identifying the unique intestinal microbiota biomarkers associated with different Tradi-tional Chinese Medicine TCM syndrome.Methods Ten one-month-old SPF-grade male Kunming mice were randomly divided into a normal group and a model group,with five mice in each group.Mice in normal group were raised regularly,and mice in model group were fed with Folium senna intragastric administration,centrifuge tube restraint and long tail clamp for 7 days to establish a Ganqichengpi di-arrhea syndrome mouse model.After successful modeling,intestinal contents were collected,and the bacterial characteristics of the intes-tinal contents were analyzed by the second generation high-throughput sequencing technology.Results The alpha diversity indices in the model group were significantly lower than those in the normal group.Firmicutes and Bacteroidetes were the dominant phyla in the two groups,and Lactobacillus and Streptococcus were the dominant genera in the two groups.Compared with the normal group,the relative a-bundance of Bacteroidetes decreased significantly(t=2.516,P=0.036),while that of Firmicutes increased significantly(t=-2.667,P=0.028).Furthermore,the relative abundance of Lactobacillus was significantly decreased(t=2.684,P=0.028),and that of Strepto-coccus was significantly increased(t=-2.453,P=0.040).LEfSe analysis results further showed that Lactobacillus was the marker bac-teria in the normal group and Streptococcus was the marker bacteria in the model group.Conclusion The modeling of Ganqichengpi di-arrhea reduced the bacterial diversity in the intestinal contents of mice.Streptococcus may be the characteristic marker bacteria in the in-testinal contents of mice with Ganqichengpi diarrhea syndrome.
