Analysis of Resistance Level of Aegilops tauschii to Mesosulfuron-Methyl and Mutation of Its Target Gene
Indoor whole-plant bioassay was conducted to determine the resistance levels of 28 Aegilops tauschii populations collected from various regions in Henan Province to mesosulfuron-methyl.Additionally,a comparative analysis of the reported mutation sites in the target acetolactate synthetase(ALS)gene was performed between highly resistant and sensitive populations of Aegilops tauschii,so as to provide a theoretical basis for the chemical control of Aegilops tauschii.The results indicated that there were 15 populations with low resistance[2≤RI(relative resistance index)<5],7 populations with moderate resistance(5≤RI≤10),and 3 populations with high resistance(RI>10)to mesosulfuron-methyl.Among them,the population TXLZ collected from Lizhuang Village of Tongxu County showed the highest resistance to mesosulfuron-methyl with inhibitory median dose(ED50)of 126.11 g/ha and RI of 31.85.Comparative analysis of the reported mutation sites in the target ALS gene among three highly resistant populations and one sensitive population did not reveal any mutations.In summary,most Aegilops tauschii populations in Henan Province have developed resistance to mesosulfuron-methyl,and the resistance of Aegilops tauschii is not caused by mutations in the target gene sites but may be due to non-target site resistance such as metabolic processes.
