Expression and correlation analysis of long non-coding RNA bladder cancer-associated transcript 1,miR-519d,and tripartite motif protein 32 in patients with triple-negative breast cancer
Objective To determine the expression and correlation of long non-coding RNA bladder cancer-associated transcript 1(lncRNA-BLACAT1),miR-519d,and tripartite motif protein 32(TRIM32)in patients with triple-negative breast cancer.Methods Among the patients with breast cancer who received treatment in the First Hospital of Lanzhou University from June 2021 to July 2022 and had complete fol-low-up data,52 patients who were diagnosed with triple-negative breast cancer and had normal distant tis-sue≥5 cm from the edge of the cancer confirmed by pathologic examination and immunohistochemistry were included in the observation group.Fifty healthy subjects who underwent physical examinations during the same period were included in the control group.The expression of lncRNA-BLACAT1,miR-519d,and TRIM32 mRNA and protein in the peripheral blood of patients in the two groups was detected by reverse-transcription polymerase chain reaction(RT-PCR)and immunoblotting.Results Compared with the con-trol group,the level of lncRNA-BLACAT1 and TRIM32 mRNA expression in patients with triple-negative breast cancer was significantly increased.In patients with triple-negative breast cancer,the level of miR-519d expression was significantly decreased.These comparisons were all statistically significant(P<0.05).The level of TRIM32 protein expression in triple negative breast cancer group was significantly higher than the control group,and the difference was statistically significant(P<0.05).Pearson correlation analysis showed that the level of lncRNA-BLACAT1 expression in the serum of patients with triple-negative breast cancer was negatively correlated with the level of miR-519d expression(r=-0.850,P<0.05),the level of lncRNA-BLACAT1 expression was positively correlated with the level of TRIM32 expression(r=0.842,P<0.05).Conclusions In patients with triple-negative breast cancer,the level of lncRNA-BLACAT1 and TRIM32 expression is increased,while the level of miR-519d expression is significantly decreased.LncRNA-BLACAT1,miR-519d,and TRIM32 might serve as potential molecular markers in the diagnosis of triple-negative breast cancer.
