Baicalin alleviates dermatitis injury of acne rats by suppressing SDF-1/CXCR4 signaling pathway
To investigate the effect of baicalin(BA)on acne-induced dermatitis injury of acne rats through regulating stromal cell-derived factor-1/CXC chemokine receptor 4(SDF-1/CXCR)signaling pathway and its mechanism.The rat model of acne was established,and the rats were divided into control group,model group,BA groups(BA-L,BA-M,BA-H groups,intragastric administration of 25,50,100 mg·kg-1·d-1 BA)and combined groups(intragastric administration of 100 mg·kg-1·d-1 BA+intraperitoneal injection of 7 µg·kg-1·d-1 SDF-1α recombinant protein).The apparent changes of auricle skin tissue were observed and the thickness of auricle was measured.HE and Masson staining were used to observe the pathological changes and fibrosis of auricle skin;ELISA was used to detect levels of IL-1β,IL-6,IL-8 and TNF-α in serum;immunofluorescence was used to detect the expression of NLRP3 in auricle skin;Western blot was used to detect the level of SDF-1 and CXCR4 proteins in auricle skin.Compared with the control group,the auricle of the model group was red and swollen,papules and telangiectasia were obvious,the stratum corneum of the ear was thickened,inflammatory cells infiltrated and the size of sebaceous gland was increased,and the hair follicle sebaceous gland was excessively keratinized.Furthermore,rats in model group demonstrated increases in thickness of auricle skin tissue,fibrotic area,hydroxyproline content,levels of IL-1β,IL-6,IL-8,and TNF-α,positive expression rate of NLRP3,and expression of SDF-1 and CXCR4 proteins(P<0.05).BA could reverse these changes mentioned above in model rat(P<0.05),while SDF-1α recombinant protein could suppress the effect of BA-H on the above indexes(P<0.05).In conclusion,BA may alleviate dermatitis injury in acne rats by inhibiting SDF-1/CXCR4 signaling pathway.
