Chemical composition analysis and anti-cellular senescence ability of aqueous extracts of Ficus pumila L.based on UHPLC-QTOF-MS
The chemical constituents in Ficus pumila L.fruits,stems and leaves were characterized by ultra-high performance liquid chromatography-quadrupole time-of-flight-mass spectrometry(UHPLC-QTOF-MS),and an investigation was conducted on their ability to alleviate lung cellular senescence induced by cigarette smoke extract.Acetonitrile-0.1%formic acid aqueous solution was used as the mobile phase for gradient elution at a flow rate of 0.2 µL·min-1;electrospray ionization(ESI)was used with positive and negative ion scanning modes for data acquisition,and the aqueous extracts of Ficus pumila L.fruits,stems,and leaves were characterized by database comparison through Msdial and by referring to the reported literature.A Beas-2b senescence model of human bronchial epithelial cells induced by cigarette smoke extract(CSE)was used to investigate the effects of aqueous extracts of Ficus pumila L.fruits,stems,and leaves on cellular activities and senescence.The findings demonstrated that thirty-three chemical components in the aqueous extracts of Ficus pumila L.fruit,stem and leaf were characterized by combining database and literature reports.The three aqueous extracts of Ficus pumila L.showed no significant toxicity to Beas-2b cells in the appropriate concentration range.50 μg·mL-1 of the aqueous extract of Ficus pumila stems and leaves showed some alleviation of cellular senescence.The results indicated that the UHPLC-QTOF-MS method is sensitive,rapid and reliable,and can comprehensively and rapidly analyze and characterize the chemical components in the aqueous extracts of different parts of Ficus pumila L.,and Ficus pumila L.has potential anti-cellular senescence ability.
