The highland barley β-glucan restored Th17/Treg cell balance to maintain intestinal function in mice
The intestinal immune system is required to perform complex tasks, such as provide a rapid, specific immune re-sponse to defend against pathogenic microorganisms and maintain immune tolerance to intestinal contents. The maintenance of intestinal immune balance is essential for intestinal homeostasis.Ulcerative colitis leads to a continuous inflammatory response, which causes an immune imbalance in the gut.To study how the highland barley β-glucan(HBG)renewed the intestinal barrier function of mice by regulating Th17/Treg cell balance.Established a mouse model of DSS-induced colitis and orally administered HBG at different concentrations. The colonic morphology of mice was observed, and the expression levels of Occludin,Claudin-1 protein,Th17 cells,Treg cells,M1 and M2 macrophages in mice colon tissue were detected. The differentiation of Th17 and Treg cells in mice spleen was detected by flow cytometry. The above results showed that HBG could alleviate the pathological symp-toms of colitis, recover the Th17/Treg balance in mice colon and spleen tissues.HBG could restore immune homeostasis and promote intestinal barrier function of colitis mice by balancing Th17/Treg cell homeostasis. It provides a theoretical basis for the development of functional food with polysaccharide to maintain intestinal immunity.
