玉米秸秆纤维素降解菌的分离纯化与产酶工艺优化

摘要
从富含腐烂玉米秸秆的土壤中分离筛选到1株能降解纤维素的细菌K01,经形态观察和16S rDNA序列分析,初步鉴定该菌属于美洲爱文氏菌。对该菌产酶条件进行优化,结果表明在接种种龄24 h,培养液初始pH值7,接种量3%,培养时间48 h的优化条件下,酶活力达1.58 IU/mL。该菌株对玉米秸秆降解效果较好,优化条件下7 d降解率达56.1%。
Abstract
A cellulose-decomposing fungus strain is isolated from soil containing rich rotten corn straw. Based on morphology and 16Sr DNA sequence analysis, it is initially classified into the genus of Penicillium. And the optimum conditions for CMCase production are determined as follows: seed age of 24 hours, initial pH value of medium of 7, inoculum amout of 3%, culture time of 48 hours. Under the optimal conditions, the CMCase activity reaches 1.58 IU/mL. The strain K01 effectively degrades the corn straw, and the degradation rate reaches 56.1%in seven days.
