免疫磁珠净化-高效液相色谱法测定香辛料中的黄曲霉毒素

为更好地监测辣椒、花椒等香辛料中黄曲霉毒素暴露状况,建立了基于免疫磁珠净化的高效液相色谱法测定香辛料中4种黄曲霉毒素B1、B2、G1、G2.样品经乙腈-水(84+16,V+V)提取后,通过黄曲霉毒素总量免疫磁珠试剂盒净化,并结合CLOVER免疫磁珠自动工作站,显著提升了净化效率.加标回收试验结果表明,在辣椒、花椒等复杂样品基质中,4种毒素的平均回收率为86%~111%,相对标准偏差小于7%.总体而言,所建立的方法准确、灵敏,适用于调味品中4种黄曲霉毒素的同步定量检测.
Determination of Aflatoxin in Spice by High Performance Liquid Chromatography Based on Immunomagnetic Beads Purification
In order to better monitor aflatoxins exposure in spices such as cumin and chili powder,we established a high-performance liquid chromatography method based on immunomagnetic beads purification to determine four aflatoxins B1,B2,G1 and G2 in spices.Aflatoxin in the sample was extracted by acetonitrile-water(84+16,V+V),and purified by aflatoxin total immunomagnetic bead kit,combined with CLOVER immunomagnetic bead automatic workstation,which significantly improved the purification efficiency.The results showed that the average recovery rate of four kinds of toxins in five kinds of spices was between 86%~111%,and the relative standard deviation was less than 7%.In general,the established method was accurate,sensitive and suitable for simultaneous quantitative detection of four aflatoxins in spices.
