Optimization of Fermentation Technology of Functional Red Kojic Rice by Response Surface Methodology
In this study, one red kojic rice strain which was selected and preserved by our laboratory was used as experimental strain.Firstly, the significantly factors of lovastatin production by red kojic rice was screened by Plackett-burman experiment, and then the maximum response area was determined by steepest ascent experiment. Finally, the optimal process conditions for high-yield lovastatin production by red kojic rice was obtained by Box-behnken experimental design and response surface analysis. The results showed that the inoculation amount, citric acid addition and glutamic acid addition were selected as significant factors affecting the content of lovastatin through P-B experiment. The optimal process conditions optimized by response surface methodology were 19% inoculation amount,1.4% citric acid addition, and 0.9% glutamic acid addition. At these conditions, the lovastatin content of functional red kojic rice reached to 12.23 mg/g.
