Isolation,identification and fungicides screening of the pathogen of tea plant anthracnose in Xiapu County of Fujian Province
Tea plant anthracnose is an important factor affecting the yield and quality of tea in Xiapu County,Fujian Province.Identifying the pathogen responsible for the disease is conducive to the targeted control.The pathogens were isolated and purified from Fuyun 6 tea tree disease leaves in Xiapu County,Fujian Province by tissue separation and polygenic molecular identification,and the fungicide inhibition rate was determined.The results showed that:there were grayish white spots on the surface of the diseased leaves,and the brown bound-ary line between sickness and health parts was clear.There were many acervulus on the spots.After cultured on PDAY medium for 4 d,the pathogen produced a colony with a diameter of 4.0 cm,with dense,fluffy and grayish-white mycelia on the surface of the colony,orange pigment at the back with punctured melanin deposi-tion.After the removal of aerial hyphae and illumination,the colonies can produce orange,oily conidial clus-ters.The conidia are close to cylindrical in shape,straight in the middle and semicircular at both ends.They lack septum,and may form black appressoria on the hydrophobic surface.The pathogen was identified as Colle-totrichum fructicola by further multigene phylogenetic tree constructed based on ITS,ACT,CAL and GAPDH gene sequences.The stress tests of different fungicides showed that the inhibition effect of chlorothalonil and pyrazolium were good,whereas carbendazim had no inhibitory effect.
