Determination of organic acids in oil tea root tissue and root exudation with HPLC
A simple and efficient method was developed for the determination of organic acids in Camellia oleifera root tissue, and the samples cultured in sand with different phosphorus concentration were determined. The root exudation was collected by the solution culture method and the organic acids were extracted from the dry root powder with ultrasonic extraction in a water bath. The correlation coefficient, recoveries, coefficient variations and limit of detection of the method was 0.999 0-0.999 9,82.3 %-108.4%,0.72 %-3.46%,1-193 μg/L,respectively,suggesting the 10 organic acids could be determined simultaneously and quickly by the method. The 10 organic acids of the samples determined by the method. The results showed that the content of organic acids in root tissue and root exudation under no phosphorus stress were higher than that under phosphorus stress,it means that the phosphorus stress encourage the organic acids accumulate in camellia's roots and secret into rhizosphere.
