日粮添加蚕豆皮对湖羊空肠屏障相关基因表达、消化酶活性和微生物菌群的影响

Effects of dietary broad bean skin on barrier-related gene expression,digestive enzyme activity and microflora of jejunum in Hu sheep

[目的]本试验旨在研究日粮添加蚕豆皮对湖羊空肠屏障相关基因表达、消化酶活性和微生物菌群的影响.[方法]30 只 4 月龄体重相近[(27±2.0)kg]的雄性湖羊分为 2 组,分别饲喂基础日粮(对照组,CON)和添加 30%蚕豆皮日粮(蚕豆皮组,BBS).预饲期 10 d,正式期 50 d.试验结束后采集空肠组织及食糜(n=5),通过RT-qPCR检测黏膜屏障相关基因的表达,生化试剂盒检测消化酶活性,高通量测序分析食糜微生物菌群.[结果]与对照组相比,BBS组空肠黏膜细胞连接基因Claudin-1、Occludin、MUC-2和ZO-1,促炎因子IL-6、IL-10 和TNF-α mRNA水平无显著变化(P>0.05),而IL-1β mRNA水平显著降低(P<0.05).2 组空肠黏膜 α-淀粉酶、胰蛋白酶和脂肪酶活性无显著变化(P>0.05).BBS 组食糜微生物Shannon、Chao1 和Observed otus指数显著降低(P<0.05);门水平上,BBS组变形菌门(Proteobacteria)相对丰度显著升高(P<0.05),厚壁菌门(Firmicutes)、髌骨细菌门(Patescibacteria)相对丰度和F/B值显著降低(P<0.05);属水平上,BBS组克里斯滕森氏菌R7 菌群(Christensenellaceae R-7 group)、未分类厚壁菌属(Firmicutes unclassified)、糖单胞菌属(Candidatus Saccharimonas)、糖酵解菌属(Saccharofermentans)、毛螺科 NK3A20 菌群(Lachnospiraceae NK3A20 group)和聚乙酸菌属(Acetitomaculum)的相对丰度显著下降(P<0.05).16S rDNA基因组的PICRUSt 2 功能预测结果显示,空肠微生物差异菌群主要富集在三羧酸循环、脂肪酸β氧化、原儿茶酸降解和L-亮氨酸降解通路.[结论]日粮添加 30%蚕豆皮可降低空肠黏膜炎症因子IL-1β mRNA表达量,改变空肠食糜微生物菌群多样性和相对丰度,调节肠道发酵环境.研究结果为蚕豆皮在养羊业发展中的应用提供参考.
[Objectives]This study aimed to investigate the effect of adding broad bean skin(BBS)to the diet on the barrier-related gene expression,digestive enzyme activity and microflora of the jejunum in Hu sheep.[Methods]Thirty 4-month-old male Hu sheep with similar body weight[(27±2.0)kg]were divided into two groups,feeding with basal diet(CON)and diet with 30%broad bean skin(BBS),respectively.The pre-feeding period was 10 days,and the formal period was 50 days.At the end of the experiment,jejunal tissue and chyme were collected(n=5),mucosal barrier-related gene expressions were detected by RT-qPCR,mucosal digestive enzyme activities were detected by biochemical kits,and chyme microbial was analyzed by high-throughput sequencing.[Results]Compared with CON group,the jejunal mucosal cell junction genes Claudin-1,Occludin,MUC-2 and ZO-1,pro-inflammatory factors IL-6,IL-10 and TNF-α mRNA levels in the BBS group had no significant changes(P>0.05),while the expression level of IL-1β mRNA significantly decreased(P<0.05).There was no significant change in the mucosal α-amylase,trypsin and lipase activities between two groups(P>0.05).The microbial Shannon index,Chao1 index and Observed otus index in the BBS group significantly decreased(P<0.05).At the phylum level,the relative abundance of Proteobacteria in the BBS group significantly increased(P<0.05),while the relative abundance of Firmicutes,Patescibacteria and F/B value significantly decreased(P<0.05).At the genus level,the relative abundance of Christensenellaceae R-7 group,Firmicutes unclassified,Candidatus Saccharimonas,Saccharofermentans,Lachnospiraceae NK3A20 group and Acetitomaculum significantly decreased(P<0.05).The functions of jejunal microbial differential flora were mainly enriched in the tricarboxylic acid cycle,fatty acid β-oxidation,protocatechuic acid degradation and L-leucine degradation pathways.[Conclusions]The addition of 30%BBS in the diet can decrease the jejunum mucosal inflammatory factor IL-1β mRNA expression level,change the jejunum chyme microbial flora diversity and relative abundance,and regulate intestinal fermentation environment.The results will provide a reference for the application of broad bean skin in the development of sheep industry.
