紫薇带芽茎段的离体培养及快速增殖

以花瓣深粉红色的紫薇品种'彩霞满天'(Lagerstroemia indica'Caixia Mantian')的半木质化带芽茎段为外植体,从消毒灭菌、初代培养基、增殖培养基等方面进行离体培养体系的优化.结果表明:各处理中紫薇茎段最佳消毒方案为先用75%酒精浸泡18 s,再用5%NaClO溶液浸泡8 min;腋芽茎段最佳启动培养基为1/2MS+6-BA 1.0 mg·L-1+NAA 0.1 mg·L-1+蔗糖20 g·L-1+琼脂6.3 g·L-1,该培养基的平均诱导率达到45%,芽长势最好且叶色嫩绿;以基本培养基、6-BA浓度、KT浓度3个因素进行L9(33)正交试验,得出影响紫薇组培苗有效增殖系数的三因素主次程度为:6-BA>基本培养基>KT;对紫薇组培苗增殖及生长效果最好的培养基为 MS+NAA 0.05 mg·L-1+6-BA 2.0 mg·L-1+KT 4.0 mg·L-1+蔗糖 30 g·L-1+琼脂 6.3 g·L-1 和WPM+NAA 0.05 mg·L-1+6-BA 2.0 mg·L-1+KT 4.0 mg·L-1+蔗糖 30 g·L-1+琼脂 6.3 g·L-1.
In vitro Culture and Rapid Propagation of Stem Segments with Buds of Lagerstroemia indica
The semi-lignified stem segments with buds of Lagerstroemia indica'Caixia Mantian'with dark pink petals were used as explants to optimize the in vitro culture system from the aspects of dis-infection and sterilization,primary medium and proliferation medium.The results showed that the ap-propriate disinfection method for stem segments of L.indica was to first soak them in 75%alcohol for 18 s,and then soak them in 5%NaClO solution for 8 min.The optimal initiation medium for stem segment with axillary buds was 1/2MS+6-BA 1.0 mg·L-1+NAA 0.1 mg·L-1+sucrose 20 g·L-1+agar 6.3 g·L-1,with an average induction rate reaching 45%,and then the buds had the best growth and the leaves was tender green in color.The L9(33)orthogonal experiment design was carried out with three factors:basic medium,6-BA concentration and KT concentration.The main and secondary factors affecting the effective proliferation coeffi-cient of L.indica tissue culture seedlings were found to be:6-BA>basic medium>KT.The culture media with the best effect on the proliferation and growth of the L.indica seedlings were MS+NAA 0.05 mg·L-1+6-BA 2.0 mg·L-1+KT 4.0 mg·L-1+sucrose 30 g·L-1+agar 6.3 g·L-1 and WPM+NAA 0.05 mg·L-1+6-BA 2.0 mg·L-1+KT 4.0 mg·L-1+sucrose 30 g·L-1+agar 6.3 g·L-1.
