Crystallization and Preliminary X-ray Diffraction Analysis of the PDZ Domain from PDLIM2
PDLIM2 (Mystique/SLIM) protein is a member of the actinin-associated LIM protein (ALP) subfamily of proteins, which contain a N-terminal PDZ domain and a C-terminal LIM domain. Studies have shown that PDLIM2 acts as an ubiquitin E3 ligase targeting nuclear p65, STAT (signal transducers and activators of transcription) proteins for proteasomal degradation, and promotes Tax oncoprotein polyubiquitination. The PDZ domain of PDLIM2 (PDLIM2-PDZ) has been reported to be a protein-protein recognition module involved in protein complex assembly. However, to date, the structure of the native PDZ domain of PDLIM2 has not been released. In this study, the PDZ domain (82 amino-acid residues) of PDLIM2 was expressed, purified and crystallized using the sitting-drop vapour-diffusion method. X-ray diffraction data were collected to 0. 173 nm resolution in-house. The crystal belongs to space group P212121, with unit-cell parameters a = 3. 820 2 nm, b=5. 091 1 nm, c=7. 777 5 nm. α=β=γ=90°. The value of the Matthews coefficient is 2.14×10-3 nm3 Da-1 for two molecules per asymmetric unit, corresponding to a solvent content of 42. 57%.
