Rosa26-LEM4定点转基因小鼠模型构建

为研究LEM4基因在肿瘤发生发展中的生物学功能,采用CRISPR/Cas9技术,在小鼠基因组的Rosa26位点插入 CAG-loxP-STOP-loxP-hLEM4-3xFLAG-IRES-EGFP,建立了Rosa26-LEM4 定点转基因小鼠模型.依据小鼠Rosa26位点序列设计合成gRNA、构建含同源序列和目的序列CAG-loxP-STOP-loxP-hLEM4-3xFLAG-IRES-EGFP的插入序列,和Cas9核酸酶mRNA共同注射入小鼠的受精卵中并获得敲入小鼠后代,经Southern blot鉴定、以及PCR扩增插入序列后的序列分析鉴定出F0首建鼠,F0首建鼠与野生型小鼠杂交后获得基因型为Rosa26-LEM4flox-LSL/wt的F1 代小鼠.Rosa26-LEM4flox-LSL/wt小鼠自交后获得 Rosa26-LEM4flox-LSL/flox-LSL基因型小鼠.Rosa26-LEM4floxLSL/flox-LSL小鼠与EIIa-cre工具鼠杂交后获得表达人LEM4蛋白的Cre+/-;Rosa26-LEM4flox-LSL/wt小鼠模型.该模型的建立为研究LEM4的肿瘤生物学功能提供了材料和思路.
Construction of Rosa26-LEM4 Site-Specific Transgenic Mice Model
LEM4 overexpression renders ER+breast cancer cells resistant to tamoxifen.However,very little is known about the function of LEM4 gene in tumorigenesis and development.CRISPR/Cas9 technol-ogy was used to knock-in hLEM4 gene at ROSA26 locus in mice,and the inserted fragment structure was CAG-Loxp-STOP-Loxp-hLEM4-3xFLAG-IRES-EGFP.Rosa26-LEM4flox LSL/wt recombinant mice were determined by Southern blot.Rosa26-LEM4flox LSL/floxLSL mice were crossed with EⅡa-cre mouse,and the Cre+/-;Rosa26-LEM4eox LSL/wt recombinant mice were identified by PCR genotyping and Western Blot-ting.Finally,the Cre+/-;Rosa26-LEM4flox LSL/wt mice model was obtained successfully.
