Impacts of Poric Acid on Epithelial Mesenchymal Transition and Immune Escape in Renal Cancer Cells
786-O cells were randomly separated into blank group,Poric acid group,CXCL12 group,CXCL12-CXCR4 inhibitor-AMD3100 group,Poric acid+CXCL12 group,and poric acid+AMD3100 group.CCK-8 was applied to detect cell proliferation.Transwell was applied to detect cell migration and invasion.Immunofluorescence was applied to detect the expression of EMT related factors such as E-Cadherin,zinc finger transcription factor(Snail),and N-cadherin.After co-culturing with CD8+T cells,Elisa was applied to detect the levels of immune escape related factors-interleukin-4(IL-4),IL-10,and interferon-γ(IFN-γ)in the supernatant.Western blot was applied to detect the expression of CXCL12,CXCR4,CXCR7,and immune escape protein PD-L1.To investigate the impacts of poric acid on epithelial mesenchymal transition(EMT)and immune escape in renal cancer cells by regulating the CXCL12-CXCR4/CXCR7 signaling pathway.Results showed that compared with the blank group,the proliferation rate,numbers of migration and invasion,the Snail,N-Cadherin IOD values,the level of IL-10,the expression of CXCL12,CXCR4,CXCR7,and PD-L1 in the Poria acid group were greatly reduced,the E-Cadherin IOD value,and the levels of IFN-γ and IL-4 were greatly increased(P<0.05).Compared with the poria acid group,the proliferation rate,numbers of migration and invasion,the Snail,N-Cadherin IOD values,the level of IL-10,the expression of CXCL12,CXCR4,CXCR7,and PD-L1 in the poria acid+CXCL12 group were greatly increased,the E-Cadherin IOD value,and the levels of IFN-γ and IL-4 were greatly reduced(P<0.05).Compared with the CXCL12 group,the proliferation rate,numbers of migration and invasion,the Snail,N-Cadherin IOD values,the level of IL-10,the expression of CXCL12,CXCR4,CXCR7,and PD-L1 were greatly reduced in the Poric acid+CXCL12 group,the E-Cadherin IOD value,and the levels of IFN-γ and IL-4 were greatly increased(P<0.05).Compared with the Poria acid group and AMD3100 group,the proliferation rate,numbers of migration and invasion,the Snail,N-Cadherin IOD values,the level of IL-10,the expression of CXCL12,CXCR4,CXCR7,and PD-L1 in the poria acid+AMD3100 group were greatly reduced,the E-Cadherin IOD value,and the levels of IFN-γ and IL-4 were greatly increased(P<0.05).The results showed poric acid can inhibit immune escape and EMT in renal cancer cells by regulating the CXCL12-CXCR4/CXCR7 signaling pathway.
