Overexpression of Msx2 Promoted the Transition of Mouse Embryonic Stem Cells to Trophoblast Stem Cells
In the first cell fate decision,identical blastomere gradually differentiate into inner cell mass(ICM)and trophectoderm(TE)two different cell lineages.However,in mice although ICM and TE originally derived from the same blastomere,they have an impassable barrier,and the lineages of ICM and TE cannot convert to each other under natural conditions.In this study,piggyBac transposons were used to construct an over-expression(OE)system of Msx2 in mouse embryonic stem cells(ESCs).And Msx2-OE mESCs were induced in the conditioned medium(CM)of trophoblast stem cells(TSCs),which can be induced transit from mESCs to mTSCs.This result provides a reference for studying the potential mechanism of the first cell fate decision.
