拟南芥中的NTMC2T4是含N端跨膜区和C2结构域蛋白家族中的一员,有研究指出在逆境条件下,NTMC2T4缺失突变体比野生型更抗旱、更耐盐,但其具体功能机制还未有报道.为了深入研究NTMC2T4,课题组纯化了 NTMC2T4胞浆段,并制备了相应的多克隆抗体.随后利用抗体进行免疫印迹和免疫共沉淀实验,发现NTMC2T4抗体可以检测到内源和外源NTMC2T4的表达.实验结果证明了 NTMC2T4抗体的特异性,为后续将要开展的NTMC2T4功能机制以及与其他蛋白互作的研究奠定了重要基础.
Antibody Preparation and Specific Detection of NTMC2T4 in Arabidopsis
NTMC2T4 is a member of the N-terminal transmembrane region and C2 domain protein family in Arabidopsis.Studies have shown that NTMC2T4 defective mutants are more resistant to drought and salt than wild type under stress conditions,but the mechanism of function has not been reported.In order to further study NTMC2T4,the research team purified the NTMC2T4 cytoplasmic segment and pre-pared the corresponding polyclonal antibodies.Immunoblotting and co-immunoprecipitation experiments were performed using antibodies.It was found that NTMC2T4 antibody can detect the expression of endogenous and exogenous NTMC2T4.The experimental results proved the specificity of the NTMC2T4 antibody,which lays an important foundation for the future research on the functional mechanism of NTMC2T4 and its interaction with other proteins.
