摘要
应用中医传承辅助平台挖掘清代医家用药规律,发现药物组合和新处方,应用网络药理学技术对新处方进行药理学机制的分析,并用糖尿病大鼠模型进行验证.研究发现,清代医家用药以补虚药、收涩药为主.对新处方"双苓泽丹方"进行网络药理学分析,得出主要成分为kaempferol、catechin、quercetin等,关键靶点有AKT1、TP53、TNF、IL6、JUN、VEGFA、CASP3、IL1B等,主要涉及到IL-17、PI3K-Akt、Relaxin等通路.动物实验显示双苓泽丹方组血糖下降,进水量、排泄量下降,血清空腹胰岛素、胰岛素抵抗指数、糖化血红蛋白和TNF-a较模型组改善.ALT、AST、ALP和UREA较模型组较低,提示双苓泽丹方可能通过改善胰岛素抵抗调控糖尿病大鼠的血糖,且有一定的肝肾保护功能.
Abstract
Based on data mining analysis,this study verifies the rule and mechanism of physicians in the Qing Dynasty in treating diabetes.TCMIAP was used to explorer the rule of physicians in the Qing Dynasty,discover drug combinations and potential new prescriptions.Network pharmacology technology was applied to analyze the mechanism of new prescriptions,which was verified with the model of diabetes rats.The research found that physicians in the Qing Dynasty often used tonic deficiency medicine and astringent medicine.A network pharmacology analysis was conducted on the new prescription"SLZDF",and the main components were identified as Kaempferol,catechin,quercetin,etc.Key targets include AKT1,TP53,TNF,IL6,JUN,VEGFA,CASP3,IL1B,etc.mainly pathways such as IL-17,PI3K Akt,and Relaxin.Animal experiments showed that in the treatment group,FBG,water intake,excretion,FINS,HOMA-IR,HbA1c,and TNF-a,ALT,AST,ALP,UREA decreased.
