Effect of Salidroside on the Blood-Brain Barrier in Rats with Acute Cerebral Infarction by Regulating the MLCK/MLC Signaling Pathway
A rat model of acute cerebral infarction was established based on the thread occlusion method,and the successfully established cerebral infarction rats were included in the model group,different Sal in-terventions(50 mg/kg,200 mg/kg)group,MLCK inhibitor(ML-7)group(1 mg/kg),and 200 mg/kg Sal+ML-7 group,respectively,the 13 rats were only separated from the common carotid artery and internal and external carotid arteries as the sham surgery group.Subsequently,neurological function scores and brain water content were measured on the rats.Evans blue(EB)was used to detect blood-brain barrier permeability.TTC was used to detect infarct volume.The apoptosis of rat brain tissue was detected by the kit.HE was applied to detect pathological changes in brain tissue.QRT-PCR was applied to detect the expression of brain tissue related junction proteins occludin and Claudin-5.Western blot was applied to detect the expression of MLCK/MLC pathway related proteins in brain tissue.The effect of Sal on blood-brain barrier in rats with acute cerebral infarction by regulating MLCK/MLC signaling pathway was investigated.The results show that compared with the sham surgery group,most neurons in the model group showed shrinkage and disappearance of nucleoli,the neurological function score,brain water content,EB exudate,cerebral infarction volume,apoptosis rate,p-MLC2/MLC2,and MLCK expression were greatly increased,the mRNA expression of Occludin and Claudin-5 was greatly reduced(P<0.05).Compared with the model group,the pathological damage in the 50 mg/kg Sal group,200 mg/kg Sal group,and ML-7 group was improved,the neurological function score,brain water content,EB exudate,cerebral infarction volume,apoptosis rate,p-MLC2/MLC2,and MLCK expression were greatly reduced,the mRNA expression of Occludin and Claudin-5 was greatly increased,and there was a statistical difference among different doses of Sal(P<0.05).Compared with the 200 mg/kg Sal group and ML-7 group,the neurological function score,brain water content,EB exudate,cerebral infarction volume,apoptosis rate,p-MLC2/MLC2,and MLCK expression in the 200 mg/kg Sal+ML-7 group were greatly reduced,the mRNA expression of Occludin and Claudin-5 was greatly increased(P<0.05).The results showed Sal improves the blood-brain barrier and protects neurological function in rats with acute cerebral infarction by regulating the MLCK/MLC signaling pathway.
