Vip3Aa诱导云杉卷叶蛾中肠CF-203细胞凋亡的研究

Vip3Aa可以进入草地贪夜蛾(Spodoptera frugiperda)的卵巢细胞(Sf9细胞)并诱导凋亡.然而,目前尚不清楚Vip3Aa激活毒素(actVip3Aa)是否能对昆虫中肠细胞产生同样的作用.本研究发现actVip3Aa可以进入云杉卷叶蛾(Choristoneura fumiferana)中肠细胞(CF-203细胞)并最终运输到溶酶体中,并且actVip3Aa进入细胞的数量与毒性呈正相关.actVip3Aa会引起CF-203细胞溶酶体pH上升、线粒体膜电势下降、基因组DNA断裂、caspase-3活性上升等现象,说明actVip3Aa能够诱导CF-203细胞凋亡.联合使用多杀菌素(spinosad)和actVip3Aa能够协同杀伤CF-203细胞,显著提高细胞的死亡率.
Vip3Aa Induces Apoptosis in Choristoneura Fumiferana Midgut CF-203 Cells
Vip3Aa can enter the ovary cells of Spodoptera frugiperda(Sf9 cells)and induce apoptosis.However,it is unclear whether Vip3Aa activated toxin(actVip3Aa)exerts the same effects on insect midgut cells.In this study,actVip3Aa was found to be able to internalize into Choristoneura fumiferana midgut cells(CF-203)and fuse with lysosomes,and the internalization level was related to its virulence.Moreover,actVip3Aa treatment could induce increased ly-sosomal pH,decreased mitochondrial membrane potential,genomic DNA fragment,and increased caspase-3 activity.These results suggested that actVip3Aa could induce apoptosis in CF-203 cells.The combined use of spinosad and actVip3Aa significantly increased the mortality of CF-203 cells,implying that spinosad and actVip3Aa had synergistic effects on killing insect cells.
