Vip3Aa Induces Apoptosis in Choristoneura Fumiferana Midgut CF-203 Cells
Vip3Aa can enter the ovary cells of Spodoptera frugiperda(Sf9 cells)and induce apoptosis. However,it is unclear whether Vip3Aa activated toxin(actVip3Aa)exerts the same effects on insect midgut cells. In this study,actVip3Aa was found to be able to internalize into Choristoneura fumiferana midgut cells(CF-203)and fuse with lysosomes, and the internalization level was related to its virulence.Moreover,actVip3Aa treatment could induce increased ly-sosomal pH,decreased mitochondrial membrane potential, genomic DNA fragment, and increased caspase-3 activity. These results suggested that actVip3Aa could induce apoptosis in CF-203 cells. The combined use of spinosad and actVip3Aa significantly increased the mortality of CF-203 cells, implying that spinosad and actVip3Aa had synergistic effects on killing insect cells.
