Isolation,Identification and Nitrogen Removal Characteristics of A Bacteria Strain with High-Efficient Aerobic Denitrifying Ability from Tianjin Coastal Wetland
The resources and functional characteristics of bacteria with high-performance nitrogen removal ability in coastal wetlands were explored in this study.A strain of aerobic denitrifying bacteria WH9 was isolated from the soil in Tianjin coastal wetland, and then it was identified as Klebsiella sp.WH9,using 16S rRNA sequence analysis. The growth and nitrogen removal performance of this strain was examined under conditions of different carbon sources, temperatures, initial nitrogen mass concentrations, and inoculum amount. The growth and nitrogen removal performance of Klebsiella sp.WH9 was significantly affected by carbon source,tem-perature,initial nitrogen mass concentration, and inoculum amount,respectively(P<0.05).The single-factor experiments showed that, the growth and total nitrogen removal efficiency of Klebsiella sp.WH9 were the highest when sucrose was the carbon source(TN removal efficiency was 90.85%),increased with the increase of temperature(TN removal efficiency was 89.72%under 35 ℃)and initial nitrogen concentration(TN removal efficiency was 86.31%under initial nitrogen concentrations of 200 mg/L),and increased at first and then decreased with the increase of inoculum amount(TN removal efficiency was 87.92%under inoculum amount of 5%).Klebsiella sp.WH9 exhibited excellent aerobic denitrification ability, and this strain has promising potential for treating nitrogen pollution in coastal wetlands.
