Objective:To investigate the effect of knockdown of NIPBL gene on TGF-β1 and IGF-1 combined to induce chondrogenic differentiation of bone marrow mesenchymal stem cells.Methods:Lentiviral-transfected NIPBL shRNA BMSCs were divided into TGF-β 1 group,TGF-β 1+IGF-1 group,NIPBL-blank control group,and an untransfected NIPBL+control group,and the above four groups of cells were subjected to chondrogenic differentiation induction.The protein expression levels of Sox-9 and Collagen-Ⅱ were detected by Western blot technique at 7,14 and 21 days of culture.Results:(1)At each stage of chondrogenesis induction,the protein expression of Sox-9 and Collagen-Ⅱ were higher in the TGF-β1+IGF-1 group than in the TGF-β 1 group,and higher in the NIPBL+control group than in the TGF-β 1 and NIPBLcontrol groups(P<0.05).(2)At 7 and 21 days of induction,the protein expression of Sox-9 gene was higher in the TGF-β 1+IGF-1 group than in the NIPBL-control group(P<0.05).(3)At each induction stage,there was no statistical difference between the TGF-β 1+IGF-1 and the NIPBL+control group.Conclusion:Knockdown of NIPBL gene TGF-β 1 combined with IGF-1 has reduced ability to induce chondrogenic differentiation of bone marrow mesenchymal stem cells.
