摘要
目的:通过生物信息学方法分析泛癌中葡萄糖-6-磷酸脱氨酶2(GlcN6P deaminase 2,GNPDA2)表达水平及免疫相关性;浅显分析肝细胞癌中(Hepatocellular Carcinoma,HCC)DNA甲基化与GNPDA2相关性.方法:利用ULCAN网站分析泛肿瘤中GNPDA2表达水平与生存差异;运用Spearman相关性分析GNPDA2表达与免疫细胞浸润及免疫分子相关性;预测DNA甲基化调节GNPDA2表达能力.结果:HCC中存在GNPDA2表达升高(P<0.001),其过表达可缩短患者生存期(P<0.01);GNPDA2过表达与免疫有关分子存在一定相关性(r>0.1,P<0.05),并促进免疫细胞浸润(r>0.2,P<0.05);DNA低甲基化可上调GNPDA2表达(r=-0.26,P<0.001).结论:HCC中GNPDA2过表达可促进免疫细胞浸润并与DNA甲基化呈负相关.
Abstract
Objective:To analyze the expression level and immune correlation of glucose-6-phosphate deaminase 2(GNPDA2)in pan-cancer by bioinformatics methods.Superficial analysis of DNA methylation and GNPDA2 correlation in Hepatocellular Carcinoma(HCC).Methods:GNPDA2 in pan-tumors was analyzed in terms of expression levels and survival differences using the ULCAN website.Spearman correlation analysis was applied to correlate GNPDA2 expression with immune cell infiltration,immune checkpoints and immunomodulatory molecules.The ability of DNA methylation to regulate GNPDA2 expression was predicted.Results:Elevated GNPDA2 expression was present in HCC(P<0.001).and its overexpression shortened patient survival(P<0.01).GNPDA2 overexpression correlated with immune-related molecules(r>0.1,P<0.05)and promoted immune cell infiltration(r>0.2,P<0.05).DNA hypomethylation upregulated GNPDA2 expression(r=-0.26,P<0.001).Conclusion:GNPDA2 overexpression in HCC promoted immune cell infiltration and was negatively correlated with DNA methylation.
