The Study on Correlation between Cytological Development Period of Astragalus Microspore and the Morphology of Floral Organ
Using Astragalus membranaceus(Fisch.)Bge.var.mongolicus(Bge.)Hsiao(AM)and Astragalus membranaceus(Fisch.)Bge(AG)as experimental materials,their cytology of AM microspore development stages,as well as the relationship between the morphological characteristics of microspore buds and anthers at different development stages were studied with acetic acid magen-ta staining and conventional paraffin sectioning method.The results showed that the development of microspores in both AM and AG undergoes the stages of microspore mother cell,tetrad,monokary-otic,and maturation,but there are significant differences in microspore characteristics among differ-ent developmental stages.The developmental stage of Astragalus microspores is closely related to the morphology of flower buds and anthers.When AM microspores are in the monokaryotic stage,the length and width of flower buds are 3.19-6.94 mm and 1.63-2.03 mm,the length of sepals is 3.17-5.91 mm,and the length and width of anthers are 0.66-0.95 mm and 0.47-0.65 mm re-spectively.The flower buds and anthers of AG during the monokaryotic stage are smaller than those of AM,with the length and width of the flower buds are 2.97-4.73 mm and 1.46-1.98 mm,the length of the sepals is 2.96-4.69 mm,and the length and width of the anthers are 0.44-0.75 mm and 0.46-0.54 mm respectively.The anther colors of AM and AG during the monokaryotic stage are both yellow.Our study provides a scientific basis for the selection of explants during in vitro cul-ture of Astragalus microspores,laying the foundation for haploid breeding and germplasm innova-tion of Astragalus.
