摘要
论文考察了稀土离子对枯草芽孢杆菌1.108液态发酵产木聚糖酶的影响,以期选育高产木聚糖酶的枯草芽孢杆菌.结果表明,于0~6 h时添加1×10-4 mol/L的Er3+,30℃液态发酵12 h,木聚糖酶活达到133.79 U/mL,获得一株木聚糖酶高产菌Bacillus subtilis X-Er,与原始菌株相比提高10.48%;连续传代培养8次,木聚糖酶活力没有显著变化,表明菌株具有良好的遗传稳定性;电镜结果发现Er3+的加入使该菌细胞壁的通透性增大,推测这可能是木聚糖酶活增加的原因之一.
Abstract
Different types of rare earth ions were added with varying concentrations in the cultivation process of Bacillus subtilis 1.108 for breeding high-yield xylanase strain.After addition 10-4 M Er3+at 0~6 h,the xylanase activity achieved 133.79 U/mL,which was 10.48%higher than the original strain.A high xylanase production strain was screened and named Bacillus subtilis X-Er,which showed good genetic stability of the high-yield xylanase during eight generations.Er3+increased cell permeability,which was one of the reasons for the increased xylanase activity.
