摘要

本研究采用高通量测序技术测定了不同林龄、不同土层油松(Pinus tabuliformis)人工林地细菌多样性和物种组成,并以附近的半固定沙地为对照,分析了科尔沁沙地油松人工林土壤细菌群落结构特征.结果表明:放线菌门(35.62%)、变形菌门(21.87%)和酸杆菌门(14.46%)是油松人工林地和对照的优势门;营造油松人工林显著改变了沙地土壤细菌群落结构(R=0.581,P=0.001);不同林龄间细菌群落结构无显著差异,但0~2 cm、2~10 cm和 10~20 cm土层间存在显著差异;油松人工林地 0~2 cm土层中的拟杆菌门较 2~10 cm和 10~20 cm提高 223.37%和 142.33%;0~2 cm土层中的黏球菌门较2~10 cm和10~20 cm提高 76.17%和39.88%;0~2 cm土层的游动放线菌属丰度较 2~10 cm和10~20 cm土层分别提高 5.70 倍和 6.69 倍;0~2 cm土层的芽单胞菌属丰度较2~10 cm和10~20 cm土层分别提高 2.22 倍和 5.09 倍.科尔沁沙地营造油松人工林显著改变了沙地细菌群落结构,林地表层土壤参与碳、氮循环及抗真菌细菌类群明显提高,促进 0~2 cm土层养分积累的同时,也提高了 2~10 cm和 10~20 cm土层养分循环的联动作用.

Abstract

High-throughput sequencing technology was used to determine the bacterial diversity and species composi-tion of Pinus tabuliformis forest with different ages and different soil layers,and the soil bacterial community structure of Pinus tabuliformis forest in Horqin Sandy Land were analyzed in this study.The results showed that Actinobacteriota(35.62%),Proteobacteria(21.87%)and Acidobaceriota(14.46%)were the dominant phyla in sample area.The construction of Pinus tabuliformis forest significantly changed the soil bacterial community structure on sandy land(R=0.581,P=0.001).There was no significant difference in bacterial community structure among different age groups,and the differences were mainly reflected in 0~2 cm,2~10 cm and 10~20 cm soil layers.The relatively abundance of Bacteroidata in 0~2 cm soil layer were increased by 223.37%and 142.33%compared with 2~10 cm and 10~20 cm,respectively.The relatively abundance of Myxococcota in 0~2 cm soil layer were increased by 76.17%and 39.88%compared with 2~10 cm and 10~20 cm,respectively.The relatively abundance of Actinoplanes in 0~2 cm soil layer was 5.70 times and 6.69 times higher than that of 2~10 cm and 10~20 cm,respectively.The relatively abundance of Gemmatimonas in 0~2 cm soil layer was 2.22 times and 5.09 times higher than that of 2~10 cm and 10~20 cm,respectively.The construction of Pinus tabuliformis forest in Horqin Sandy land significantly changed the bacterial community structure,and the participation of carbon and nitrogen cycling and anti-fungal bacterial groups in the forest surface soil significantly increased,which promoted the accumulation of nutrients in 0~2 cm soil layer,and also improved the interaction of nutrient cycling in 2~10 cm soil layer and 10~20 cm soil layer.
