基于LC-MS/MS技术的4种柳属植物叶片的差异代谢物分析

为探究腹毛柳(Salix delavayana)、坡柳(Salix myrtillacea)、康定柳(Salix paraplesia)和乌柳(Salix cheilophila)叶片代谢物差异,本文采用液相色谱串联质谱技术分析4种柳属植物叶片代谢物成分,并结合主成分分析(PCA)和正交偏最小二乘法(OPLS-DA)筛选差异代谢物,基于KEGG分析差异代谢物的富集通路.结果发现:在4种柳属植物叶片中共检测到19类2 796个化合物,主要为异戊二烯类、黄酮类、苯及其取代衍生物、羧酸及其衍生物、有机氧和脂质化合物;腹毛柳与坡柳、康定柳、乌柳间,差异代谢物分别有641个、625个、643个,坡柳与康定柳、乌柳,以及康定柳与乌柳间,差异代谢物分别有592个、593个、626个,差异代谢物主要以黄酮类为主;4种柳属植物叶片的差异代谢物主要富集于黄酮和黄酮醇生物合成、类黄酮生物合成、ABC转运蛋白等途径上.研究结果可为这4种柳属植物资源的综合利用提供参考,并为其他柳属植物间的差异代谢物分析提供了研究新思路.
Analysis of Differential Metabolites in Leaves of Four Salix Plants Based on LC-MS/MS Technology
This study aimed to investigate the differential metabolites in the leaves of Salix delavayana,Salix myrtillacea,Salix para-plesia,and Salix cheilophila.The Liquid chromatography-tandem mass spectrometry(LC-MS/MS)technology was used to analyze the metabolites in the leaves of four species of Salix plants.Principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to identify differential metabolites,and KEGG analysis was conducted to determine the enriched pathways of these metabolites.The result showed that a total of 2 796 compounds belonging to 19 classes were detected in the leaves of four Salix plants,mainly including isoprene,flavonoids,benzene and its substituted derivatives,carboxylic acids and their derivatives,organic oxygen,and lipid compounds.There were 641,625,and 643 differential metabolites between Salix dela-vayana and Salix myrtillacea,and between Salix delavayana and Salix paraplesia,and between Salix delavayana and Salix cheiloph-ila,respectively.Additionally,592,593,and 626 differential metabolites were found between Salix myrtillaceae and Salix para-phylesia,and between Salix myrtillacea and Salix cheilophila,and between Salix paraplesia and Salix cheilophila.The differential metabolites were mainly flavonoids.The differential metabolites in the leaves of four Salix plants mainly enriched in pathways such as flavonoid and flavonol biosynthesis,flavonoid biosynthesis,and ABC transporter proteins.The findings provided a reference for the comprehensive utilization of these four species of Salix plant resources,and offered new insights into differential metabolite analy-sis among other Salix plants.
