Study on the Mechanism of Modified Qiyu Ointment in Promoting Wound Repair based on NetworkPharmacology
Objective:To explore the potential mechanism of modified Qiyu ointment in promoting wound repair based on network pharma-cology.Methods:The active ingredients of each traditional Chinese medicine component in the modified Qiyu ointment prescription were screened through the TCMSP database,and the potential targets of the active ingredients were predicted using the SwissTargetPrediction data-base;the Genecards database was used to search for"wound repair"(WoundRepair)related targets;use Cytoscape 3.7.1 software to build a"drug-component-target"network,with the targets being drug-disease intersection genes;build a protein interaction network(PPI)of drug-disease intersection targets through the STRING database to obtain the key targets of Qiyu ointment modified prescription to promote wound repair;Use the DAVID database to enrich and analyze GO functions and KEGG signaling pathways on key targets,and build a drug-target-channel-disease network.Results:76 active ingredients,216 drug targets,and 1478 wound repair targets were screened out,and 118 drug-disease intersection targets were finally obtained.The PPI network data was further screened to obtain 35 genes such as AKT1,TP53,IL6,TNF,JUN,CASP3,EGFR,PTGS2,IL1B,and MMP9 as core targets;GO enrichment analysis identified 404 biological processes,29 cell com-ponents,and 52 molecular functions.KEGG signaling pathway enrichment analysis involved 133 signaling pathways.Conclusion:The active ingredients of the modified Qiyu ointment may play a role in quercetin,kaempferol,β-sitosterol,formononetin,and isorhamnetin.Various ac-tive ingredients may act on AKT1,TP53,IL6,TNF,JUN,CASP3,EGFR,PTGS2,IL1B,and Atherosclerosis,AGE-RAGE signaling pathway in diabetic complications,human cytomegalovirus infection,endocrine resistance,TNF signaling pathway,proteoglycans in cancer and other pathways.Multiple core targets such as MMP9 play various functions such as regulating the expression of inflammatory factors such as IL6,TNF,PTGS2,and IL1B,stimulating new capillary formation,inducing fibroblast migration,and regulating cell proliferation and differentiation to interfere in various processes of wound healing and promote wound repair,but the results need further experimental verification.
