Influence of Shearing Mode and Humiture on Proliferation Effect of Cerasus humilis cv.Four Plantlets in Vitro
Cerasus humilis cv.Four plantlets will occur seedling aging phenomenon constantly when subcultured on a same formula medium many times,fully utilize the specificity of germinated shoots from axillary buds which Cerasus humilis cv.Four stem segment plantlets subcultured.Share off the upper part stem segment,then take protective measure on the remaining cutting seedlings,including reserve 2 axillary buds from downslope on upslope when inoculated and sheared,and then subcultured on the culture shelf which set the humidity and temperature are 65~73%and 29±1℃respectively.The axillary buds can germinate bright and shaped shoots,the number of shoots is above 2.8,the length of shoots is above 4.69 cm,other measurement indicators such as the leaf area,the content of chlorophyll and the leaf specific weight represent excellent performance.Furthermore,there is no apparent seedling aging and etiolation.Through adjusting factors of shearing mode and humiture,mitigate the trend of seedling aging,and supply eligible inoculated plantlets for next subculture.
