Research on the Therapeutic Effect of Icariin on Testicular Function Injury Induced by Busulfan in Male Mice
Objective To investigate the therapeutic effect of icariin(ICA)on testicular function impairment induced by busulfan in male mice.Methods Molding state:mice were divided into a control group(20%DM-SO)and a model group(4 mg·kg-1 busulfan),with 8 mice in each group.ICA treatment of model mice were di-vided into 4 groups,with 11 mice in each group:control group(20%DMSO),model group(4 mg·kg-1 busulfan),ICA treatment group(4 mg·kg-1 busulfan+120 mg·kg-1 ICA),lycium barbarum polysaccharide(LBP,serve as positive comparison)treatment group(4 mg·kg-1 busulfan+40 mg·kg-1 LBP).All experimental mice were 6-week-old ICR male mice.The weight of the testicles and epididymis were measured.A cow abalone style counting board for sperm counting was used,automated sperm analyzer was used to detect sperm survival rate,and Diff-Quik detection kit was employed to detect sperm deformity rate.HE stained testic-ular tissue were observed,and Johnsen score was evaluated.Malondialdehyde(MDA),glutathione(GSH)and lipid peroxides(LPO)contents in testicular tissue were measured by corresponding assay kits,and the mito-chondrial morphology in spermatogonia were observed through transmission electron microscopy.Results Compared with the control group,the weight of the testicles and epididymis of the model group by busulfan de-creased(P all<0.05),the testicular index decreased(P<0.01),the number of sperm and sperm survival rate sig-nificantly decreased(P all<0.001),and the total sperm deformity rate significantly increased(P<0.001),the Johnsen score significantly decreased(P<0.001),the content of MDA in testicular tissue were significantly in-creased(P<0.01),and the content of GSH were significantly decreased(P<0.01),LPO content were significantly increased(P<0.001).Compared with the model group,the weight of the testicles and epididymis,testicular in-dex,sperm survival rate,and Johnsen score of the ICA treatment group were all increased(P all<0.001),the number of sperm increased(P<0.01),and the total sperm deformity rate decreased(P<0.001),the content of MDA in testicular tissue were decreased(P<0.01),the content of GSH were increased(P<0.001),and the con-tent of LPO were decreased(P<0.001).Conclusion ICA showed significant therapeutic effects on busulfan induced impairment of testicular function in male mice.
