Objective To analyze the expression level of X-ray repair cross complementary protein-5(XR-CC5)in brain glioma(BG)and its relationship with clinicopathological features and prognosis.Methods The expression difference of XRCC5 in 636 BG[468 cases of low-grade glioma(LGG),168 cases of glioblastoma multiforme(GBM)]and normal brain tissues were analyzed using RNA sequencing data from the Cancer Gene Atlas(TCGA)database.The expression level of XRCC5 was correlated with age,sex,pathological type and grade,survival time and expression level of tumor marker of proliferation Ki-67(MKI67).XRCC5 immunohistochem-ical staining was performed on BG tissue chips(47 cases LGG,75 cases GBM and 3 cases normal human brain tissues)to further verify the relationship between the expression level of XRCC5 and the pathological classifi-cation of BG.Results The results of TCGA analysis indicated that the expression level of XRCC5 in BG tis-sues was significantly higher than that in normal brain tissues(P<0.001),and the expression level in the GBM group was significantly higher than the LGG group(P<0.001).The expression level of XRCC5 was positively correlated with the pathologic grade of BG(P<0.001)and the expression level of MKI67(P<0.001).The sur-vival period of the high expression group was longer than that of the low expression group(P<0.001).There was no significant difference in XRCC5 expression level between different sex and age groups(P all>0.05).Immuno-histochemical staining of XRCC5 indicated that the expression level of XRCC5 in tumor tissue was signifi-cantly higher than that in normal brain tissue(P<0.05).Conclusion XRCC5 was significantly elevated in glioma tissue and positively correlated with MKI67,a biomarker of malignant tumor.The higher the expres-sion of XRCC5,the better the prognosis and the longer the survival.
