摘要
目的 明确胃癌细胞系AGS中活性氧(ROS)产生的水平,并探讨通过调节ROS含量对人胃癌细胞系AGS增殖、迁移、侵袭的影响及其机制.方法 将胃癌细胞系AGS分为ROS清除对照组、ROS清除(1∶500)组、ROS清除(1∶200)组,ROS诱导对照组、ROS诱导(1∶1 000)组、ROS诱导(1∶500)组;DHE荧光探针检测各组胃癌细胞AGS中ROS的水平;Transwell、细胞划痕实验法检测各组细胞的侵袭和迁移能力;Western blot实验检测不同水平的ROS对P53的调控影响.结果 与对照组相比,ROS清除组细胞的增殖侵袭、迁移能力增强(P<0.01),且ROS清除剂浓度越高胃癌细胞的增殖、侵袭、迁移能力越强(P<0.01);与对照组相比,ROS诱导组细胞的增殖、侵袭、迁移能力降低(P<0.05),且ROS诱导剂浓度越高细胞的增殖、侵袭、迁移能力下降越明显(P<0.01);高水平的ROS上调P53蛋白的表达(P<0.01),低水平的ROS下调P53蛋白的表达(P<0.O1).结论 高水平ROS抑制胃癌细胞增殖、侵袭、迁移能力,其可能通过调控P53而发挥相应作用.
Abstract
Objective To determine the level of ROS production in gastric cancer cell line AGS,and explore the effect of regulating ROS content on the proliferation,migration and invasion of gastric cancer cell line AGS and its mechanism.Methods The gastric cancer cell line AGS was grouped into ROS clearing control group,ROS clearing group(concentration 1∶500),ROS clearing group(concentration 1∶200),ROS induction control group,ROS induction group(concentration 1∶1 000),and ROS induction group(concentration 1∶500).DHE fluorescent probe was used to detect the level of ROS in AGS of gastric cancer cells in each group.Transwell,cell scratch assay were applied for invasion and migration of each group.Western blot experiment was used to detect the effect of different levels of ROS on P53.Results Compared with the control group,the proliferation,invasion and migration of cells in ROS scavenging group were enhanced(P<0.01),and the proliferation,inva-sion and migration of gastric cancer cells were enhanced with the higher concentration of ROS scavenging agent(P<0.01).Compared with the control group,the proliferation,invasion and migration of cells in ROS induced group were decreased(P<0.05),and the proliferation,invasion and migration of cells in ROS induced group were significantly decreased with the higher concentration of ROS inducer(P<0.01).High ROS levels up-regu-lated P53 protein expression(P<0.01),low ROS levels down-regulated P53 protein expression(P<0.01).Con-clusion High levels of ROS inhibit the proliferation,invasion and migration of gastric cancer cells,and ROS may play a corresponding role by regulating P53.
