Early Prevention and Treatment of Lycium Barbarum Polysaccharide on C57BL/6J Osteoporosis Mice
Objective To discuss the early prevention and treatment effect of Lycium barbarum polysaccharide(LBP)on C57BL/6J osteoporosis mice.Methods 12 weeks C57BL/6J female mice were selected to make osteoporosis model by bilateral ovariectomy.In the sham group,fat tissue around the ovaries was removed,then the successful model mice were divided into model group and LBP group 1 week after surgery,LBP group was given LBP 0.1 g·kg-1,once a day for 12 weeks,sham group and model group were given the same amount of distilled water.The content of estradiol(E2)in serum of mice was detected by ELISA.Serum oxidative stress indexes malondialdehyde(MDA)and superoxide dismutase(SOD)were detected by biochemical methods.The Micro CT scan of tibia was performed to observe the changes of trabecular thickness(Tb·Th),ratio of bone surface area to bone volume(BS/BV),trabecular separation(Tb·Sp),bone density(BMD),bone volume fraction(BV/TV)and bone volume(BV).Results Compared with sham group,the growth rate of body mass in model group was increased,uterine coefficient,serum E2 content were decreased,serum oxidative stress index MDA content were increased,SOD activity were decreased(P all<0.01).Compared with model group,there was no significant difference in uterine coefficient and serum E2 content in LBP group(P all>0.05),but the growth rate of body mass,serum MDA content were decreased,SOD activity was increased(P all<0.01),bone BMD and BV were increased(P all<0.05),Tb·Th and BV/TV were significantly increased(P all<0.01).The three dimensional reconstruction of tibia showed that the cortex bone in the model group was thin,osteoporosis was present,the number of trabeculae bone was reduced,the arrangement was disordered,and the pulp cavity was obviously enlarged.The number of trabeculae bone in LBP group was higher than that in model group.Conclusion LBP may play a role in ameliorating osteoporosis by regulating oxidative stress response in early osteoporosis mice.
