西黄丸溶出度测定及不同制丸方法比较

目的 建立西黄丸溶出度测定方法,比较泛制法与塑制法 2 种生产工艺溶出曲线的相似性,对泛制法与塑制法工艺样品进行一致性评价.方法 通过筛选 4种溶出介质、3种转速及2种测定方法,最终确定pH 1.2盐酸为溶出介质,转速为 100 r·min-1,溶出方法为小杯法,溶出样品和介质的体积比为 3 g/200 mL;同时,分别测定西黄丸泛制法与塑制法中胆红素的溶出度,采用非模型依赖法中的差异因子(f1)、相似因子(f2)对溶出曲线的相似性进行评价.结果 在 90 min时,产品的溶出率达到最高;按确定的溶出试验参数,以胆红素含量为评价指标进行精密度、重复性、稳定性等方法学考察,结果均符合要求;且 2 种生产工艺样品中胆红素溶出曲线的相似度良好.结论 泛制法与塑制法生产工艺对产品质量无明显影响,产品质量具有一致性.
Determination of Dissolution in Xihuang Pills and Comparison of Different Pelleting Methods
Objective To establish methods for determining the dissolution rate of Xihuang pills,compare the similarity of dissolution curves between the pan-made method and plastic-made method,and evaluate the consistency of samples produced by the pan-made method and plastic-made method.Methods By screening four kinds of dissolution media,three kinds of rotational speed and two kinds of determination methods,pH 1.2 hydrochloric acid was finally determined as the dissolution medium,the dissolution speed was 100 r·min-1,the dissolution method was the small cup method,and the volume ratio of the dissolution sample to the dissolution medium was 3 g/200 mL.At the same time,the dissolution degree of bilirubin in Xihuang pill from the pan made method and plastic-made method was determined,and the similarity of dissolution curve was evaluated by difference factor(f1)and similarity factor(f2)in model-independent method.Results The dissolution rate reached the highest at 90 min.According to the determined dissolution test parameters,the precision,repeatability and stability of bilirubin all met the requirements.The dissolution degree of bilirubin in the two production processes was similar.Conclusion The quality of the products produced by the pan-made method process and plastic-made method process is no obvious influence,and the quality of the products is consistent.
