摘要

目的 探讨喹硫平对匹罗卡品致痫大鼠海马组织中基质金属蛋白酶-9(MMP-9)的表达及认知功能的影响.方法 建立氯化锂-匹罗卡品癫痫模型,随机分为正常对照组与癫痫组,其中癫痫组分为生理盐水组和喹硫平组.通过IntelliCage全自动监测系统观察大鼠认知功能损害情况;应用免疫组织化学染色法、Western blot法检测大鼠海马组织中MMP-9 的表达情况.结果 行为学实验研究显示,癫痫组大鼠的认知功能低于正常对照组大鼠,喹硫平组大鼠的认知功能高于生理盐水组(P均<0.05).分子生物学实验研究显示,癫痫组大鼠海马组织中MMP-9 的表达量高于正常对照组大鼠,喹硫平组大鼠海马组织中MMP-9 的表达量低于生理盐水组(P均<0.05).结论 喹硫平能降低癫痫大鼠海马组织中MMP-9 的表达,并改善其认知功能.

Abstract

Objective To investigate whether the quetiapine has effects on the cognitive function and expression of matrix metalloproteinase-9(MMP-9)in hippocampus of epileptic rats.Methods A lithium chloride-pilocarpine-induced epilepsy rats'model was established,and rats were randomly divided into the normal control group and the epilepsy group.And the epilepsy group was further divided into the normal saline group and the quetiapine group.The cognitive function of rats was observed by IntelliCage automatic monitoring system.The expression of MMP-9 in hippocampus was detected by immunohistochemistry and Western blot.Results Behavioral studies showed that the cognitive function of epilepsy rats was significantly lower than the normal control rats.In the epilepsy group,the cognitive function of the quetiapine group was higher than the normal saline group(P all<0.05).The results of molecular biology experiments showed that the expression level of MMP-9 in hippocampus of the epilepsy group was significantly higher than that of normal control group.The expression level of MMP-9 in hippocampus of the quetiapine group was lower than the normal saline group(P all<0.05).Conclusion Quetiapine could reduce the expression of MMP-9 in the hippocampus of epileptic rats and improve the cognitive function.
