The Medication Rules and Mechanism of Prescriptions for Treating Eczema Based on Traditional Chinese Medicine Inheritance Assistance Platform and Network Pharmacology
Objective By exploring traditional Chinese medicine formulas for treating eczema and utilizing network pharmacology to reveal the effective ingredients of core drug groups or optimal drug pairs,and clarifying the interactions between these ingredients and targets related to treating eczema,to provide scientific basis for the treatment of eczema sing traditional Chinese medicine approaches.Methods Firstly,relevant traditional Chinese medicine formulas for treating eczema were retrieved from the"Dictionary of Traditional Chinese Medicine Formulas".The detailed information of these formulas was then entered into the Traditional Chinese Medicine Heritage Support Platform System(V2.5).The entered formula information was subjected to association rule analysis and high-frequency combination analysis.Through these analyses,complementary relationships between medicines and high-frequency drug combinations used in eczema treatment can be unveiled.Subsequently,databases such as TCMSP were utilized to perform ingredient analysis on the high-frequency drug combinations.This analysis helps to identify the effective active constituents and their corresponding target points.Tools like OMIM and GeneCards were employed to gather gene target information related to eczema,contributing to the construction of a relevant target database.Based on this database,protein-protein interaction(PPI)analysis was conducted,revealing the interplay among these target points.Finally,GO functional enrichment analysis was carried out to comprehend the functional categorization of eczema-related target points,further unveiling their roles in the occurrence and development of the disease.Utilizing the KEGG database,pathway enrichment analysis was performed,aiding in the determination of the biological pathways involving eczema-related target points.This comprehensive approach leads to a deeper understanding of the mechanisms of treatment.Results The study has collected information on 104 formulas,encompassing 181 different types of Chinese herbs.Among these,20 herbs appeared with a frequency of 10 or more times.The high-frequency herbal combinations were associated with a total of 361 drug target points and 644 disease-related target points,with a common interaction involving 46 targets.Through PPI analysis,key proteins crucial to the process of eczema treatment by traditional Chinese medicine have been identified.Notably,proteins such as STAT1,MAPK1,and AKT1 are believed to play pivotal roles in treating eczema.These proteins are implicated in 20 cell biological processes,12 cell components,and 15 molecular functions.Pathway enrichment analysis revealed 11 enriched signaling pathways,including the AGE-RAGE signaling pathway,TNF signaling pathway,Chemical cancer receptor activation signaling pathway,B cell receptor signaling pathway,NF-κB signaling pathway,Fc γ receptor mediated phagocytic signaling pathway,chemokine signaling pathway,etc.Conclusion The prescription for treating eczema is mainly composed of heat clearing and dampness reducing drugs,supplemented by blood cooling and diuretic drugs.Medications for treating eczema have the characteristics of multiple targets,pathways,and effects,and may exert therapeutic effects by affecting angiogenesis,inflammatory factors,immune regulation,and other pathways.
