The Effect of Inducing Inflammatory Response after Epileptic Seizures on Learning and Memory as Well as the Positive Expression of Erk1/2,p-Erk1/2 and Caspase-3 in Epileptic Rats
Objective To study the changes of hippocampal Erk1/2,p-Erk1/2 and Caspase-3 expressions in epileptic SD rats at different periods.Methods The epileptic rats'model was successfully induced by intraperitoneal injection of lithium chloride-pilocarpine.The learning and memory of the rats in each group were analyzed by positioning cruise experiment.The expression of Erk1/2,p-Erk1/2 and Caspase-3 proteins in each group were determined by Western blot analysis.Results The results of positioning cruise experiment showed that the epileptic rats were much longer in the time and the total distance of positioning cruise in epileptic rats(P all<0.05).Western blot analysis results showed that the protein level of Tuj-1 expression in epileptic rats was significantly decreased(P all<0.001).The protein levels of GFAP and Iba1 expression in SE 1 d and SE 7 d groups were evidently increased(P all<0.01).The expression levels of Erk1/2 proteins in the SE 7 d,14 d and SE 28 d groups were significantly decreased(P all<0.01).The expression levels of p-Erk1/2 proteins in the SE 1 d and SE 28 d groups were significantly increased(P all<0.05).The protein levels of Caspase-3 expression in each group were also markedly up-regulated(P all<0.05).Conclusion Neuroinflammatory response can be induced after epileptic seizure in SD rats,the learning memory abilities of epileptic rats were decreased.Erk1/2 signaling pathway is activated after epileptic seizure,which induced a cascade of apoptosis,astrocytes were abnormally activated,finally a large number of neurons died,and the expression of Caspase-3 protein was significantly up-regulated.
