Study on the Establishment of Hyperuricemia Model in Rats by Fructose Combined with Potassium Oxonate
Objective The hyperuricemia(HUA)model of rats was established by fructose combined with potassiumoxonate to provide reference for the establishment and drug development of HUA model.Methods The HUA model in rats was established with 5%fructose combined with 100 mg·(kg·d)-1 potassium oxonate solution.After 4 weeks of modeling,the indicators of uric acid(UA),liver function(ALT,AST),kidney function(BUN,Scr),blood lipids(TC,TG,HDL-C,LDL-C)and fasting blood glucose(FPG)were observed.Lasso regression was used to screen variables and multiple linear regression was used to analyze variables.After 4 weeks,the experimental rats stopped consuming 5%fructose water and was randomly divided into a potassium oxonate group and a non-potassium oxonate group observe the recovery of uric acid,liver and kidney function and blood lipids at week 8,as well as the effects of potassium oxonate on various indicators.Results The results of experiment showed that at the end of the 4th week,the feed consumption of experimental rats was lower than that of control rats,while the fructose water intake,ALT,AST,and Scr were higher than those of control rats(P all<0.05).The variables selected by Lasso regression were ALT,BUN,and Scr,while the equation for multiple linear regression analysis was YUA=-383.717+0.676×ALT+31.551 x BUN+10.589×Scr.After stopping drinking fructose water,the feed consumption of potassium oxonate group in rats was significantly higher than that of control rats at the 6th week,and the BUN was higher than that of control rats and non-potassium oxonate group in rats(P all<0.05).At week 8,the water intake of non-potassium oxonate group in rats was higher than that of control rats(P<0.05).There were no statistical significance in liver and kidney function,blood lipids and other indicators(P all>0.05).Conclusion The HUA model in rats was successfully established by 5%fructose combined with 100 mg·(kg·d)-1 potassium oxonate with liver and kidney functions damaged in some extent.
