High Expression of HNF4α Promotes Cell Proliferation,Migration,and Poor Prognosis in Lung Adenocarcinoma
Objective To study the expression pattern of the HNF4α gene in lung adenocarcinoma(LUAD)and its impact on patient prognosis,as well as to explore the biological functions of HNF4α in LUAD cells,particularly its effects on cell proliferation and migration.Methods Based on TCGA and GTEx database,the expression pattern of the HNF4 α gene and its relationship with patient prognosis investigated by analyzing LUAD data.Kaplan-Meier survival analysis was utilized to evaluate the correlation were between HNF4αexpression and patient survival time,and gene set enrichment analysis(GSE A)was used to reveal the biological pathways associated with HNF4 α.Furthermore,the HNF4α overexpressing LUAD cell line was established by using lentivirus infection,and the overexpression efficiency of HNF4 α was verified by western blot andRT-qPCR experiments.The effect of HNF4 α on the proliferation and migration of LUAD cell was verified by CCK-8 and wound healing assays respectively.Results HNF4 α was significantly upregulated in LUAD and was associated with shorter overall survival(OS),progression-free survival(PFS),and disease-free interval(PFI)(P all<0.05).GSEA analysis revealed that HNF4α was closely linked to key biological processes in LUAD.Western blot and RT-qPCR experiments showed that the HNF4α overexpressing cell line was successfully constructed.CCK-8 experiment showed that HNF4α overexpression significantly enhanced the proliferation capabilities of A549 cell,and wound healing experiment showed that HNF4 α overexpression promoted the migration of A549 cell(P all<0.05).Conclusion HNF4 α is highly expressed in LUAD and is significantly related to poor prognosis,and overexpression of HNF4α significantly promotes the proliferation and migration of LUAD cells.HNF4α may serve as a potential prognostic marker and therapeutic target for LUAD.
