Expression and Clinical Significance of Silenct Information Regulator 1 and Hypoxia Inducible Factor-1α in Cutaneous Malignant Melanoma
Objective To investigate the expression of SIRT1 and HIF-1 α in cutaneous malignant melanoma(CMM)and analyze their relationship with clinical factors.Methods A total of 30 surgical specimens of CMM and melanocytic nevus confirmed by histopathology and immuno-histochemistry were collected from the Department of Dermatology of General Hospital of Ningxia Medical University from February 2022 to February 2024.The expressions of SIRT1 and HIF-1 α were detected by RT-qPCR and immunohistochemical method in CMM and melanocytic nevus tissue samples.Chi-square test was used to analyze and evaluate the correlation between the expression of SIRT1 and HIF-1 α and the gender,age,tumor site,tumor lesion diameter and pathological grade of patients.Results RT-qPCR results showed that the relative expression level of SIRT1 mRNA in melanocytic nevus was lower than that of CMM(P<0.01).The relative expression level of HIF-lα mRNA in melanocytic nevus was lower than that of CMM(P<0.05).Immunohistochemical experiment results showed that the expression of SIRT1 in CMM was significant high than that in melanocytic nevus(t=10.14,P<0.001),and the expression of HIF-1α increased in CMM,which was significant high than that in melanocytic nevus(t=21.99,P<0.001).The expression intensity of SIRT1 was correlated with tumor Clark grade and lymph node metastasis(P all<0.05).The expression intensity of HIF-1 α was correlated with tumor grade,site and lymph node metastasis(P all<0.05).Conclusion The expression of SIRT1 and HIF-1 α in CMM tissues were increased,and they were related to Clark grade,lymph node metastasis,and tumour site,which could be used as biomarks to predict tumor invasion,metastasis and prognosis.
