目的 研究通心络胶囊对糖尿病大鼠心功能及心肌重塑的影响.方法 选取Spargue Dawley(SD)雄性大鼠83只,用2%链脲佐菌素(STZ)30mg/kg造模后,将其随机分为通心络高、中、低剂量组、缬沙坦组、糖尿病组.各组灌胃10周;监测其体重及血糖;大鼠麻醉后测定其心功能,测定心脏质量指数,ELISA法测定大鼠心肌匀浆TGF-β1、Caspase-3、bcl-2蛋白含量.结果 通心络胶囊的高、中、低剂量组及缬沙坦组较糖尿病组精神状态更活跃,毛色有光泽,多食、多饮、多尿症状减轻,大便较干燥,体重增加.造模后,通心络高、中、低剂量组、缬沙坦组及糖尿病组较空白对照组明显下降,灌胃4~10周,各组较空白对照组差异均有统计学意义(P<0.05).血糖比较:灌胃1~10周,通心络高、中、低剂量组、缬沙坦组及糖尿病组较空白对照组指标显著升高(P<0.05),糖尿病组和其余实验组比较差异无统计学意义(P>0.05).心室间隔厚度、室后壁厚度、室舒张末期内径、左室收缩末期内径、左室舒张末期容积、左室收缩末期容积指标比较,缬沙坦组和通心络高剂量组较糖尿病组各指标均明显降低(P<0.05).射血分数和短轴缩短率指标比较,各组指标较糖尿病组均明显升高(P<0.05);通心络高剂量组较缬沙坦组射血分数明显增高(P<0.05).缬沙坦组和通心络高剂量组的心脏质量指数及左心室质量指数较糖尿病组明显减低(P<0.05).TGF-β1和Caspase-3指标中,缬沙坦组和通心络高、中剂量组较糖尿病组均明显降低(P<0.05),缬沙坦组和通心络高剂量组间差异无统计学意义(P>0.05).bcl-2指标中,缬沙坦和通心络高、中剂量组较糖尿病组明显增加(P<0.05),通心络高剂量组明显高于缬沙坦组(P<0.05).结论 通心络胶囊可以明显改善糖尿病大鼠的心功能、抑制心肌重塑,疗效可观.
Effects of Tongxinluo capsule on cardiac function and myocardial remodeling in diabetes rats
Objective To study the effects of Tongxinluo capsule on cardiac function and myocardial remodeling in diabetes rats.Methods Eighty three male SD rats were randomly divided into high,middle and low dose of Tongxinluo groups,valsartan group and diabetes group after being modeled with 2%STZ(30 mg/kg).The treatment group was gavaged for 10 weeks and the general condi-tion of the rats was observed;Body weight and blood sugar were monitored;Cardiac function was also measured after anesthesia in rats,and cardiac mass index was measured after heart extraction.The expression of TGFβ1,Caspase-3 and bcl-2 proteins were measured by using ELISA.Results General situation:The high,medium and low dose groups of Tongxinluo capsule and valsartan groups were more active in mental state,shiny in hair color,and the symptoms of overeating,drinking and urination are alleviated,the stool was dry,and the body mass was increased compared with the diabetes group.Weight comparison:After modeling,Tongxinluo high,medium and low dose groups,valsartan group and diabetes group significantly decreased compared with the blank control group.At 4-10 weeks of intragastric administration,the difference between each group and the control group was statistically significant(P<0.05).Blood glucose compari-son:at 1-10 weeks after intragastric administration,the indexes of Tongxinluo high,medium and low dose groups,valsartan group and di-abetes group were significantly higher than those of the control group(P<0.05),and there was no statistical significance between diabe-tes group and other experimental groups(P>0.05).Comparison of cardiac function indexes:comparison of ventricular septal thickness,posterior wall thickness,ventricular end diastolic diameter,left ventricular end systolic diameter,left ventricular end diastolic volume,and left ventricular end systolic volume,all indexes of valsartan and Tongxinluo high-dose groups were significantly lower than those of dia-betes group(P<0.05).Compared with diabetes group,ejection fraction and short axis shortening rate were significantly higher in each group(P<0.05);The high-dose group of Tongxinluo showed a significant increase in ejection fraction compared to the valsartan group(P<0.05).Comparison of cardiac mass index:the whole heart mass index and left heart mass index of valsartan group and Tongxinluo high-dose group were significantly lower than those of diabetes group(P<0.05).Comparison of protein expression of TGF-β1 and Caspase-3:The ratio of valsartan group and Tongxinluo high and middle dose groups was significantly lower than that of diabetes group(P<0.05);There was no statistically significant difference(P>0.05)between the valsartan group and the high-dose Tongxinluo group.Bcl-2 index:Valsartan and Tongxinluo high and middle dose groups were significantly higher than diabetes group(P<0.05);The high-dose group of Tongxinluo was significantly higher than the valsartan group(P<0.05).Conclusion Tongxinluo capsule can significantly improve the heart function and inhibit myocardial remodeling in diabetes rats,with considerable effect.
