The expression of TEAD1 in myocardial fibroblasts after acute myocardial infarction in mice
Objective To observe the expression of transcription factor(TEAD1)in myocardial fibroblasts after acute myocar-dial infarction in mice,and to provide a theoretical basis for exploring its mechanism of action.Methods Forty SPF grade male C57BL/6 mouse were selected and randomly divided into acute myocardial infarction group(AMI group)and sham-operation group(Sham group).After surgery,they were fed with ordinary feed until day 1,3,7,and 14,and then sacrificed and sampled.The real-time elec-trocardiogram changes and TTC staining were used to judge whether the model succeed.The expression of TEAD1 in the heart was detec-ted by Western blotting.The expression of TEAD1 in myocardial tissue was detected by double labeling of immunofluorescence and wheat germ agglutinin(WGA)staining.The expression of TEAD1 in myocardial fibroblasts was detected by double labeling of immunofluores-cence.Results Compared with Sham,the expression of TEAD1 in the heart increased significantly on the 3rd and 7th day in AMI group(P<0.05).The expression of TEAD1 in the infarct border zone increased significantly on each days in AMI group(P<0.05),showing a trend of first increasing and then decreasing,and the expression level was highest on the 3rd day.There was a statistically significant difference in the expression level of TEAD1 between adjacent days(P<0.05).The results of double labeling of immunofluorescence showed that TEAD1 was highly expressed in myocardial fibroblasts in myocardial tissue after acute myocardial infarction.Compared with Sham group,the quantity of cell expression TEAD1 in myocardial fibroblasts increased significantly after acute myocardial infarction(P<0.05),mainly expressed in the infarct border zone on 7th and 14th day.Conclusion The expression of TEAD1 in the infarct border zone of myocardial tissue increased significantly after acute myocardial infarction,and it is highly expressed in myocardial fibroblasts.
