Screening ferroptosis genes in ruptured abdominal aortic aneurysm based on random forest algorithm
Objective To explore the expression of genes related to ferroptosis in the tissues of elective abdominal aortic aneu-rysms(eAAA)and ruptured abdominal aortic aneurysm(rAAA),and to screen the characteristic genes of ferroptosis associated with rAAA,providing new insights for further revealing the molecular mechanisms of rAAA.Methods The GSE98278 dataset was download-ed from the GEO database,and the set of ferroptosis-related genes(FRGs)was obtained from the FerrDb database.Data analysis was performed by using R software version 4.1.1,and differential expression of ferroptosis-related genes(FDEGs)in eAAA and rAAA tis-sues were obtained through the Limma package.GO and KEGG functional annotation analyses of FDEGs were conducted by using the clusterProfiler package and Cytoscape's ClueGO.The importance of classification for FDEGs was determined by using the random forest algorithm,identifying the top three important genes as characteristic genes of ferroptosis in rAAA.The different expressions between eAAA and rAAA were analyzed and the diagnostic ROC curves were plotted to assess their diagnostic value for rAAA.Results Three characteristic genes of ferroptosis in rAAA were identified by using the random forest algorithm(SLC39A14,NDRG1,and TRIM21).Compared to eAAA,SLC39A14 and NDRG1 were highly expressed in the rAAA group,while TRIM21 was lower expressed in the rAAA group(P<0.05).Conclusion SLC39A14,NDRG1,and TRIM21 are characteristic genes of ferroptosis in rAAA,which may provide new ideas for research on its pathogenesis and prevention.
